Background

Belonging to the small GTPase superfamily/Ras family, RASL12 is localized in the cellular membrane and cytoplasm. RASL12 has many important molecular functions including GTP binding, GTPase activity and nucleotide binding. The main biological function of this gene is to participate in the GTP catabolic process, signal transduction and small GTPase mediated signal transduction. RASL12 interacts with ACVR1, SMAD1, SMAD2, SMAD3 and SMURF2. Documented diseases associated with RASL12 include acute kidney tubular necrosis, intraepithelial neoplasm, retinitis, endometrial cancer, coronary artery disease and Huntington's disease.

Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - An intracellular stain was performed on Jurkat cells with Alexa Fluor 647 conjugate of GPR78/HSPA5 antibody NBP1-06274AF647 (blue) and a matched isotype control NBP2-24893AF647 (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.
Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - An intracellular stain was performed on HeLa with NBP1-06274 and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550.
Immunocytochemistry/Immunofluorescence: GRP78/HSPA5 Antibody [NBP1-06274] - NIH-3T3 cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-GRP78/HSPA5 at 2 ug/mL overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

PVDF membrane was probed with 0.2 µg/mL of Goat Anti-Human Protein S/PROS1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4036) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). Specific bands were detected for Protein S/PROS1 at approximately 80 & 100 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Simple Western lane view shows human plasma, loaded at 0.2 mg/mL. A specific band was detected for Protein S/PROS1 at approximately 106 kDa (as indicated) using 2 µg/mL of Goat Anti-Human Protein S/PROS1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4036) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 700.

A specific band was detected for Catalase at approximately 64 kDa (as indicated) in the parental HeLa cell line, but is not detectable in knockout HeLa cell line. GAPDH (Catalog # AF5718) is shown as a loading control. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
