PKM2 Antibody [Biotin] Summary

Description
Store vial at 4C prior to restoration. For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

This antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Biotin, anti-Goat Serum as well as purified and partially purified Pyruvate Kinase [Rabbit Muscle]

Application Notes
This product has been tested by western blot and is suitable to be assayed against 1.0 ug of Pyruvate Kinase in a standard capture ELISA using Peroxidase Conjugated Streptavidin and ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:3,000 to 1:15,000 of the reconstitution concentration is suggested for Anti-Pyruvate Kinase Antibody.

Background

Pyruvate kinase is also known as ATP:pyruvate phosphotransferase. At least three molecular forms with pyruvate kinase activity are known. PK1, the red cell form, is deficient in a type of hemolytic anemia. PK2 is found in kidney. PK3 is found in leukocytes, muscle, platelets, and brain but not in red cells or kidney. PK1 is found also in liver. During fetal development, PK3 changes to PK1 in the liver. PK1 is a tetramer composed of two dissimilar polypeptides of somewhat different molecular weight. It is an allosteric enzyme exhibiting cooperative binding for phosphoenolpyruvate and sensitivity to fructose-1,6-diphosphate. PK3 also is a tetrameric protein but, unlike PK1, all subunits are alike and, not unexpectedly, there is no cooperative behavior. The enzyme is insensitive to fructose-1,6-diphosphate. Patients with deficiency of red cell PK have normal PK2 and PK3. There are 4 isozymes of pyruvate kinase in mammals: L, R, M1 and M2. Ab38237 recognizes the M1/M2 isoform of pyruvate kinase
