Application Notes
According to Gudipaty et al "only this Piezo1 antibody reliably worked for Piezo1 immunostaining, as it vanishes in cells mosaically expressing a Piezo1-shRNA-mCherry construct" (PMID: 28199303). Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended.

Background
Piezo1 (Mib, FAM38A) is a mechanosensitive ion channel which responds to mechanically activated currents in numerous cells, including the brain, bladder, colon, kidney, lung, and skin. However, mRNA expression of Piezo1 is weak in the DRG, unlike Piezo2. The expression of Piezo1 is induced in Alzheimer's associated senile plaques (PMID 16854388), as well as integrin activation through R-Ras signaling (PMID 20016066). R-Ras is recruited to the endoplasmic reticulum which increases calcium release, leading to the activation of the protease calpain. Piezo proteins are fairly novel proteins, and whose sequences are not akin to other ion channels or proteins.

I have read your data sheets that in WBs the bands seen are approximately 286 kDa and 318 kDa, respectively. I would greatly appreciate if you could show me these WBs on human cells For PIEZO1 and 2 and the effects of the blocking peptides (or point me to publications using your Abs).
We offer a PIEZO1 blocking peptide specific to product NBP1-78446. We unfortunately do not offer a PIEZO2 blocking peptide. Our western blot data for NBP1-78446, NBP1-78624, and NBP1-78538 was performed by an outside collaborator and they have not given permission to us to release their images. That is why we do not have them available on our website. We will fully guarantee all of our products for any listed application or species. We unfortunately do not have any publications for these products either.
