Background

Neuropilin-1 is a type I transmembrane protein that is expressed in the developing nervous system and by endothelial and tumor cells. Neuropilin-1 binds members of the class III secreted semaphorin subfamily as well as some isoforms of VEGF family proteins. The amino acid sequence of rat Neuropilin-1 extracellular domain is 98% and 93% identical to that of mouse and human Neuropilin-1, respectively.

CommentsPublished: See PMID 22927827Independent review from antibodyresource.com"Human: Immersion fixed brain tissue in 4% paraformaldehyde in 0.1 M phosphate buffered saline (PBS), pH 7.4, for three weeks at 4C, and then immersed in 0.1 M PBS containing 30% sucrose for two days at 4C.Mouse: Heads from the embryos were washed thoroughly in cold 0.1M PBS, fixed in fixative solution (4% paraformaldehyde, 2% picric acid in 0.1M PBS; pH 7.4) 6 to 8 hours at 4°C and cryoprotected in 20% sucrose overnight at 4°C. Next day, the heads were embedded in OCT (Optimal cutting temperature embedding medium, Tissue-Tek®, Sakura, France, Villeneuve d’Ascq), frozen on dry ice and stored at -80°C until for sectioning. Postnatal day 0 (PND 0) mice were anaesthetized with i.p injection of chloral hydrate and perfused transcardially with 4% paraformaldehyde, 2% picric acid in 0.1M PBS; pH 7.4. Carefully the skin was removed and the heads obtained were postfixed in the same solution for 4 hours at 4°C, cryoprotected in 20% sucrose solution overnight at 4°C. Next day, the heads were embedded in OCT, frozen on dry ice and stored at -80°C until sectioning. Cryosections (20 mm thick) werecut and processed for immunohistofluorescence.The antibody was used either at 1:400 or 1:1000dilution depending on conditions."
