Simple Western: MUC4 Antibody [NBP1-52193] - Image shows a specific band for MUC4 in 1.0 mg/mL of HeLa lysate. This experiment was performed under reducing conditions using the 12-230 kDa separation system.
In Western blot bands are seen ~78 kDa representing MUC4 beta and ~50 kDa. The identity of the 50 kDa band is unknown. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. Does not work for IP as per customer feedback.
In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue.

MUC4 (Mucin-4) is a large transmembrane protein that belongs to mucins family which contains high-molecular weight glycoproteins with oligosaccharides attached to serine or threonine residues of mucin core protein backbone by O-glycosidic linkages. Mucins have two catagories: transmembrane mucins (MUC1, MUC3, MUC4, MUC12, MUC13, MUC15, MUC16, MUC17, MUC20 and MUC21) and secreted mucins (MUC2, MUC5AC, MUC5B, MUC6, MUC7, and MUC19). Mucins are produced by epithelial cells and serve protective as well as lubricating role, however, in damaged epithelia or tumor cells, there is a loss of polarity that occurs in association with activation of proliferation/survival mechanisms. MUC4 is normally expressed in many epithelial tissues such as respiratory, colonic, and vaginal epithelia, and its expression is altered in various tumors including fibromyoxoid sarcoma and carcinomas of pancreas, bile duct, pancreatic duct etc. MUC4's mediated activation of ErbB2 downstream signaling may lead to cell proliferation/survival, and tumor cells can exploit versatile anti-apoptotic activities of MUC4 for augmenting survival or developing resistance to therapeutic agents. MUC4 has also been shown to enhance invasion and metastasis in prostate cancer, partly through regulating the EMT process (N-cadherin upregulation) and stabilizing FGFR1.
