Western blot shows lysates of MDA‑MB‑468 human breast cancer cell line, Raji human Burkitt's lymphoma cell line, and Neuro‑2A mouse neuroblastoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human/Mouse ERK5/BMK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2848) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for ERK5/BMK1 at approximately 115 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
ERK5/BMK1 was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Goat Anti-Human/Mouse ERK5/BMK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2848) at 15 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Simple Western lane view shows lysates of HeLa human cervical epithelial carcinoma cell line and NIH‑3T3 mouse embryonic fibroblast cell line, loaded at 0.2 mg/mL. A specific band was detected for p38 alpha at approximately 43 kDa (as indicated) using 5 µg/mL of Rabbit Anti-Human/Mouse/Rat p38 alpha Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8691). This experiment was conducted under reducing conditions and using the12-230 kDa separation system.
Western blot shows lysates of HEK293T human embryonic kidney parental cell line and p38 alpha knockout HEK293T cell line (KO). PVDF membrane was probed with 0.5 µg/mL of Rabbit Anti-Human/Mouse/Rat p38 alpha Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8691) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for p38 alpha at approximately 38 kDa (as indicated) in the parental HEK293T cell line, but is not detectable in knockout HEK293T cell line. 1 ug/lane of Recombinant Human BMP-4 was resolved with SDS-PAGE under reducing (R) and non-reducing (NR) conditions and visualized by silver staining, showing major bands at 22-25 kDa and 37-41 kDa, respectively. Multiple bands in gel are due to variable glycosylation. 
Recombinant Human BMP-4 (Catalog #314-BP/CF) induces alkaline phosphatase production in the ATDC5 mouse chondrogenic cell line. The ED50 for this effect is 2.5-15 ng/mL.
