Background

SUV39H1 (suppressor of variegation 3-9 homolog 1) is a histone methyltransferase that specifically trimethylates histone H3 at lys 9. Trimethylation stimulates the binding of heterochromatin proteins (HPs) which leads to epigenetic transcriptional repression in heterochromatin regions.

Western Blot: Histone H3 [Trimethyl Lys9] Antibody (6F12-H4) [NBP1-30141] - Whole cell protein from human HeLa, A431, mouse Neuro2A and rat PC12 cells was separated on a 4-20% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. Knockout Validated: HDAC1 Antibody [NB100-56340] - Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and HDAC1 knockout (KO) HeLa cell line. PVDF membrane was probed with 2.0 ug/ml of Rabbit Anti-Human HDAC1 Polyclonal Antibody (Catalog # NB100-56340) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for HDAC1 at approximately 65 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.
