• Application Notes: Neutralization: Yields one-half maximal inhibition of the biological activity of hIL-6 (1.0 ng/ml). ELISA: To detect hIL-6 by direct ELISA (using 100ul/well antibody solution) a concentration of at least 1:2000 of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hIL-6. Western Blot: Used in conjunction with compatible secondary reagents the detection limit for recombinant hIL-6 is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Background

IL-6 is a multifunctional cytokine that has an important role in host defense by regulating immune and inflammatory responses. It is expressed by a variety of normal and transformed cells including T cells, B cells, monocytes/macrophages, fibroblasts and various tumor cells. IL-6 has diverse biological functions, it stimulates B-cell differentiation and antibody production, synergizes with IL-3 in megakaryocyte development and platelet production, induces expression of hepatic acute-phase proteins, and regulates bone metabolism. Recombinant human IL-6 is a 20.9 kDa protein containing 184 amino acid residues. Human IL-6 is active on murine cells but murine IL-6 is inactive on human cells.

1. I was wondering if you know of any blocking antibodies against IL-6 or VEGF-C that can be used in cell culture to quench these cytokines?
   • We have no antibodies that are currently known to show blocking antibodies to VEGF-C. Please see this link to our IL-6 antibodies with blocking/neutralizing activity can be found at this link.
2. Could you tell me the concentration of the antibody NB600-1532. I cannot see this information on the website
   • Our lab have just confirmed for me that lot 016k4852 is at 8.7mg/ml.
3. Could you tell me how much of this antibody I will need to neutralise 90pg/ml of IL6?
