Background

Palmitoyltransferase with broad specificity. Palmitoylates GABA receptors on their gamma subunit (GABRG1, GABRG2 and GABRG3), which regulates synaptic clustering and/or cell surface stability. Palmitoylates glutamate receptors GRIA1 and GRIA2, which leads to their retention in Golgi

In the presence of a cross-linking antibody, Mouse polyHistidine Monoclonal Antibody (Catalog # MAB050) and the metabolic inhibitor actinomycin D (1 µg/mL), Recombinant Human TRAIL R1/TNFRSF10A Fc Chimera (Catalog # 347‑DR) inhibits Recombinant Human TRAIL/TNFSF10 (Catalog # 375‑TL) induced cytotoxicity in the L‑929 mouse fibroblast cell line in a dose-dependent manner (orange line), as measured by crystal violet staining. Under these conditions, inhibition of Recombinant Human TRAIL/TNFSF10 (12 ng/mL) activity elicited by Recombinant Human TRAIL R1/TNFRSF10A Fc Chimera (10 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human TRAIL R1/TNFRSF10A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF347). The ND50 is typically 0.02‑0.055 µg/mL.
Western blot shows lysates of TF‑1 human erythroleukemic cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human TRAIL R1/TNFRSF10A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF347) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for TRAIL R1/TNFRSF10A at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions.
TRAIL R1/TNFRSF10A was detected in immersion fixed paraffin-embedded sections of human brain using Goat Anti-Human TRAIL R1/TNFRSF10A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF347) at 10 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Immunohistochemistry-Paraffin: TRAILR2/TNFRSF10B Antibody [NB100-56618] - TRAIL R2/TNFRSF10B Antibody [NB100-56618] - Analysis of a formalin fixed paraffin-embedded (FFPE) human brain cerebellum using 1:200 conc. of TRAIL R2/TNFRSF10B antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 30 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 9.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 15 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Cytoplasmic staining was observed in the Purkinje cell layer.
Immunohistochemistry: TRAILR2/TNFRSF10B Antibody [NB100-56618] - Pharmacodynamic assays for ONC201. IHC analyses of CHOP, TRAILR2/TNFRSF10B, and double-stranded DNA breaks (TUNEL) in baseline and ONC201-treated biopsies for an endometrial cancer patient. Immunocytochemistry/Immunofluorescence: APE Antibody (13B8E5C2) [NB100-116] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. The cells were incubated with anti-APE (13B8E5C2) at 5 ug/mL overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
Western Blot: APE Antibody (13B8E5C2) [NB100-116] - Whole cell protein from human HeLa, A431, mouse Neuro2A and rat PC12 cells was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/mL anti-APE-1 in block buffer and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
