The GATA3-RE/LUCPorter™ reporter cell line can be used for screening of agonists, antagonists or signaling inhibitors related with the GATA3-involved signaling pathways. The cell line is designed to measure the transcriptional activity of GATA3. As a zinc-finger transcription factor, GATA3 (GATA-binding protein 3) plays a critical role in early and late T cell differentiation, which regulates Th1/Th2 differentiation. GATA3 has been shown to induce Th2 differentiation and repress Th1 differentiation. GATA3 is also known to promote the secretion of IL-4, IL-5 and IL-13 from Th2 cells.
According to the Cancer Genome Atlas Network's study, GATA3 is one of three genes (GATA3, TP53 and PIK3CA) of which somatic mutations occurred at over 10% incidence across all breast cancers. The GATA3 induction by phorbol 12-myristate 13-acetate (PMA) is shown in Figure 1. In the test, PMA induces the transcriptional activity of GATA3 in a dose response manner. GATA3 then binds to GATA3-RE leading to expression of the luciferase reporter protein, which is quantified by luciferase activity (figure 1).

Notes
