EMMPRIN/CD147 Antibody (MEM-M6/2) [CoraFluor™ 1]

CoraFluor™ 1, amine reactive (Catalog:7920) and CoraFluor™ 2, amine reactive (Catalog # 7950) are terbium-based probes that have been developed for use as TR-FRET donors. They emit wavelengths compatible with commonly used fluorescent acceptor dyes such as BODIPY® (or BDY) and Janelia Fluor® dyes, FITC (Catalog # 5440), TMR and Cyanine 5 (Catalog # 5436). CoraFluor™ fluorescence is brighter and more stable in biological media than existing TR-FRET donors, leading to enhanced sensitivity and improved data generation. CoraFluor™ 1 exhibits excitation upon exposure to a 337 nm UV laser.

EMMPRIN/CD147 Antibody (MEM-M6/2) [CoraFluor™ 1] Summary

• Description: CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(TM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

• Immunogen: A soluble recombinant form of CD147, CD147Rg, which consists of the cDNA coding for the entire extracellular region of CD147 fused to the DNA coding for the hinge region, CH2 and CH3 domain of human IgG1.
• Epitope: This antibody recognises extracellular Ig domain D1 of CD147 antigen.

• Specificity: The antibody MEM-M6/2 recognizes extracellular Ig domain D1 of CD147 (Neurothelin), a 50-60 kDa type I transmembrane glycoprotein primarily expressed on all leukocytes, red blood cells, platelets and endothelial cells; it is not expressed by resting lymphocytes. The antibody MEM-M6/1 is a high-affinity antibody capable of binding to unstimulated peripheral blood T cells.

Background

CD147 plays a pivotal role in spermatogenesis, embryo implantation, neural network formation and tumor progression. It stimulates adjacent fibroblasts to produce matrix metalloproteinases (MMPS). CD147 may target monocarboxylate transporters SLC16A1, SLC16A3 and SLC16A8 to plasma membranes of retinal pigment epithelium and neural retina. CD147 also seems to be a receptor for oligomannosidic glycans. In vitro, it promotes outgrowth of astrocytic processes. CD147 forms homooligomers in a cis-dependent manner on the plasma membrane. It forms a complex with MMP1 at the tumor cell surface. CD147 interacts with SLC16A1 and SLC1A3; probably a BSG dimer is associated with a monocarboxylate transporter dimer. CD147 also interacts with ATP1B2, MAG and L1CAM.
