This Dnmt3a antibody is useful for Immunohistochemistry on paraffin-embedded sections, Immunoprecipitation, Immunocytochemistry/Immunofluorescence and Western Blot, where a band should be seen at ~120 kDa. NOTE: This antibody does not neutralize the enzymatic activity of its corresponding methyltransferase. In ICC/IF, nuclear and cytoplasmic staining was observed in Ntera-2 cells. In IHC-P, staining was observed in the cytoplasm of mouse testis. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. With a critical role in long-term transcriptional silencing, DNA methylation represents a key layer of epigenetic information which is essential for embryonic development, germline differentiation, functional maturation of neuronal tissues etc., and DNA-methyltransferases such as DNMT3A, DNMT3B and DNMT3L are unique enzymes with the ability to transfer methyl group derived from folate intermediates to nucleic acids. DNA (cytosine-5)-methyltransferase 3A or DNMT3A is required for genome-wide de novo methylation and for establishment of DNA methylation patterns during development. Localized in the nucleus and cytoplasm, it accumulates in major satellite repeats at pericentric heterochromatin and is activated through binding to its regulatory factor DNMT3L. DNA methylation is coordinated with methylation of histones and DNMT3A modifies DNA in a non-processive manner, and it also methylates non-CpG sites. DNMT3A preferentially methylate DNA linker between 2 nucleosomal cores and is inhibited by histone H1. It plays a role in paternal/maternal imprinting and is required for methylation of most imprinted loci in germ cells. DNMT3A acts as a transcriptional corepressor for ZNF238 and actively represses transcription through HDAC.

Does this antibody recognize the short (723 aa / 81 kD) or the long (912 aa/102 kD) form of the DNA methytransferase 3A?
The immunogen sequence used for Dnmt3a antibody (NB100-265) is from the amino acids 10-118 of human Dnmt3a which does not match the amino acid sequence of the short forms of this target (i.e. 723 AA Isoform-2 or 166 AA Isoform-3). Therefore this antibody should not be able to detect the short forms of Dnmt3a. In our testing on HCT-116 cells, we have observed the target bands at approximately 120 kDa, and no bands were observed at approximately 82kD or 17kD positions.
