Specificity
It recognizes polypeptides of 58kDa and 56kDa, identified as cytokeratin 5 and 6 respectively. It shows no reaction with keratin 1, 8, or 19. It stains the basal cells of squamous and glandular epithelia, part of stratum spinosum of squamous epithelium, myoepithelium, and mesothelium. It does not stain adenous epithelium. Therefore, this antibody can be used to distinguish squamous carcinoma from adenocarcinoma, and to differentiate epitheliod mesothelioma from non-epitheliod mesothelioma and adenocarcinoma. It is also a useful antibody in the identification of mesothelioma and lung squamous cell carcinoma. It does not react with pulmonary adenocarcinoma.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Immunohistochemistry-Paraffin: RPE65 Antibody (401.8B11.3D9) [NB100-355] - Analysis of FFPE human glioblastoma tissue section using 1:500 dilution of RPE65 [401.8B11.3D9] antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems).

Immunocytochemistry/Immunofluorescence: Vimentin Antibody [NB300-223] - Analysis of HeLa cell culture. Antibody at 1:10,000 in green, and costained with mouse mAb to actin, dilution 1:500 in red. The blue is DAPI staining of nuclear DNA. The vimentin antibody stains the intermediate filament network while the actin antibody labels the submembranous cytoskeleton, stress fibers, and bundles of actin associated with cell adhesion sites.
Immunohistochemistry-Frozen: Vimentin Antibody [NB300-223] - 10 um Cryosections from PFA fixed murine brain acute slices stained with Vimentin antibody (1:600) and Alexa Fluor 488 anti chicken IgG. Antibody showed specific staining, also of blood vessels. IHC-Fr image submitted by a verified customer review.
Immunocytochemistry/Immunofluorescence: Vimentin Antibody [NB300-223] - Human A7 cells. Cells were fixed with 10% formalin and permeabilized with 0.1% TritonX-100. After blocking, cells were incubated with primary antibody (1:600) at 4C O/N followed with Alexa Fluor 488 labeled secondary antibody and counterstained with DAPI and Phalloidin for nuclei and F-actin respectively. Imaged with Leica TCS SP5 confocal microscope. ICC/IF image submitted by a verified customer review.

Flow Cytometry: p53 Antibody (PAb 240) [NB200-103] - An intracellular stain was performed on HeLa cells with p53 (PAb240) NB200-103AF647 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.
Flow Cytometry: p53 Antibody (PAb 240) [NB200-103] - An intracellular stain was performed on A549 cells with p53 (PAb240) NB200-103AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.
Flow (Intracellular): p53 Antibody (PAb 240) [NB200-103] - An intracellular stain was performed on HeLa cells with p53 (PAb240) NB200-103 (blue) and a matched isotype control NBP2-27287 (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by APC-conjugated anti-mouse IgG secondary antibody F0101B.
