Background

The protein encoded by this gene is a member of the homeodomain family of DNA binding proteins. It may regulate gene expression, morphogenesis, and differentiation and it may also play a role in the cell cycle progession. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been determined.

CommentsJurkat T cell acute lymphoblastic leukemia (T-ALL) cells were grown to late log phase, at a density of 850,000 cells per ml. Sheared chromatin was prepared from 30 million cells using the Covaris TruChIP kit and the Covaris LE220 sonicator according to the manufacturer's instructions. Sonication was performed at a 30% duty cycle for 15 min.
The chromatin was precleared with MagnaChIP Protein A + G magnetic beads (Millipore) and 10 micrograms of pure rabbit normal IgG (Jackson Immunoresearch).
The input was divided into 4 IPs of about 7 million cells each. Normal rabbit IgG (Jackson) and the 3 indicated Novus antibodies were used to IP overnight with MagnaChIP beads. The next day the beads were washed with low salt buffer, high salt buffer, LiCl buffer, and TE. The recovered DNA was eluted and purified with the iPure v2 kit (Diagenode). The recovered DNA was diluted 1:10 and 5 microliters used in a 10 microliter qPCR reaction with the QIAGEN QuantiNova SYBR Green kit.
Amplicons were from the promoters of 5 genes: INS, ALAS1, HES1, RPA3, and JAK2.
