Immunocytochemistry/Immunofluorescence: BAK Antibody (SU32-07) [NBP2-67460] - Staining Bak in CRC cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: BAK Antibody (SU32-07) [NBP2-67460] - Analysis of paraffin-embedded human small intestine tissue with Rabbit anti-Bak antibody washed with ddH2O and PBS, and then probed with the primary antibody at 1/500 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow Cytometry: BAK Antibody (SU32-07) [NBP2-67460] - Analysis of NIH/3T3 cells with Bak antibody at 1/50 dilution (blue) compared with an unlabelled control (cells without incubation with primary antibody; red). Alexa Fluor 488-conjugated goat anti rabbit IgG was used as the secondary antibody.

Immunohistochemistry-Paraffin: BAK Antibody (SU32-07) [NBP2-67460] - Immunohistochemical analysis of paraffin-embedded human stomach carcinoma tissue using anti-BAK antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (ET1608-21, 1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

Bak a pro-apoptotic analog to Bax, is a member of the Bcl-2 family of proteins. During mitochondria-regulated programmed cell death, Bak promotes apoptosis by binding Bcl-2 and inhibiting its anti-apoptotic function (1). Upon apoptotic stimulation, Bak translocates from the cytosol to the mitochondrial membrane (2). Pro-apoptotic cascade activates BID, which oligomerizes Bak into pores that result in the release of cytochrome c (2, 3). In healthy cells, Bak associate with Bcl-xL and Mcl-1 but not with Bcl-2, Bcl-w or A1 and only when freed of both Bak will induce apoptosis.
