HEK293 human embryonic kidney cell line transfected with either (A) mouse B7-H2 or (B) irrelevant transfectants and eGFP were stained with Rat Anti-Mouse B7‑H2 Monoclonal Antibody (Catalog # MAB1582) followed by Goat Anti-Rat IgG APC-conjugated Secondary Antibody (Catalog # F0113). In a functional flow cytometry test, Biotinylated Recombinant Mouse ICOS Fc Chimera (10 ng/mL) binds to HEK293 human embryonic kidney cell line transfected with mouse B7-H2. Binding is blocked by (A) 2.5 μg/mL of Rat Anti-Mouse B7-H2 Monoclonal Antibody (Catalog # MAB1582) but not by (B) Rat IgG Isotype Control (Catalog # MAB005) at 2.5 μg/mL. Cells were stained with Streptavidin-APC (Catalog # F0050).

Background

B7-H2, also called B7RP-1 and ICOSL, is a 60 kDa member of the B7 family of immune costimulatory proteins. It is expressed on resting B cells, dendritic cells, monocytes, alveolar epithelial cells, and placental trophoblasts. B7-H2 interactions with ICOS promote lymphocyte differentiation and activation. Within the ECD, mouse B7-H2 shares 49% and 70% aa sequence identity with human and rat B7-H2, respectively.
