Background

AMP-activated protein kinase (AMPK) is highly conserved from yeast to plants and animals and plays a key role in the regulation of energy balance at both the cellular and the whole body levels. Once activated, it affects a metabolic switch from an anabolic to a catabolic state, both by acutely phosphorylating metabolic enzymes and, in the longer term, by regulating gene expression. Binding APM to the beta domains triggers increased phosphorylation at Thr172 on the activation loop of the alpha subunit. AMP Phosphorylation at Thr172 is catalyzed by the tumor suppressor kinase LKB1 or CaMKK-beta, TGF-beta activated kinase-1 (TAK1).

  1. I have questions about your product, NBP1-74502 (AMPK alpha 1 p Thr172 antibody). I’d like to detect p-AMPK alpha 1 (Thr172) by Western blotting. I can see an image of Western blotting in your product sheet, wherein, the sample is from HEK293. I’d like to know how much protein is loaded in this case? Please tell me the blocking condition and what solution is used to dilute the antibody?
    • For the western of product NBP1-74502, we loaded 30ug of lysate in the gel. We blocked the blot with 5% non-fat dry milk and 1% BSA in TBST (0.1% Tween) for 1 hour at room temperature. The primary antibody was diluted in the blocking buffer.
