Background

AGER mediates interactions of advanced glycosylation end products (AGE). These are nonenzymatically glycosylated proteins which accumulate in vascular tissue in aging and at an accelerated rate in diabetes. It is receptor for amyloid beta peptide.This gene encodes a member of the immunoglobulin superfamily of cell surface molecules. It is a receptor for various molecules, including the amyloidogenic form of serum amyloid A, amyloid-beta protein, members of the S100/calgranulin superfamily and advanced glycation end products. The gene lies within the major histocompatibility complex (MHC) class III region on chromosome 6. Alternative splicing results in two transcript variants encoding different isoforms.

Western Blot: S100B Antibody (15F9NB) [NBP2-45267] - Total protein from human brain (1) and mouse brain (2) was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane for 30 minutes and blocked in 5% non-fat milk in TBST. The membrane was probed with 2 ug/ml anti-S100B in 1% milk and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: S100B Antibody (15F9NB) [NBP2-45267] - Analysis in A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with S100B (15F4NB) at a 1:100 dilution overnight at 4 degrees Celsius and detected with DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were detected with DAPI (Blue) at 2.0 ug/ml in 1X PBS. Cells were imaged using a 40X objective.

Adjacent tissue section was processed for immunohistochemistry using Mouse Monoclonal (Novus Biologicals catalog #NB100-56566) at 5ug/mL with 1 hour incubation at room temperature followed by incubation with anti-mouse IgG VisUCyte HRP Polymer Antibody (Catalog # VC001) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to lymphocytes.
