Immunocytochemistry/Immunofluorescence: PML Protein Antibody - BSA Free [NB100-59787] - NIH3T3 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with PML Protein Antibody (NB100-59787) at 2ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: PML Protein Antibody - BSA Free [NB100-59787] - A431 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with PML Protein Antibody (NB100-59787) at 1ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western blot shows recombinant human UCP1, recombinant human UCP2, recombinant human UCP3, and recombinant human UCP4 (5 ng/lane). PVDF Membrane was probed with 0.5 µg/mL of Mouse Anti-Human/Mouse UCP1 Monoclonal Antibody (Catalog # MAB6158) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 2.

Western blot shows lysates of mouse brown adipose tissue and mouse adipose tissue. PVDF Membrane was probed with 0.5 µg/mL of Mouse Anti-Human/Mouse UCP1 Monoclonal Antibody (Catalog # MAB6158) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for UCP1 at approximately 33 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 2.

UCP1 was detected in immersion fixed human mesenchymal stem cells undifferentiated (lower panel) or differentiated into adipocytes (upper panel) using Mouse Anti-Human/Mouse UCP1 Monoclonal Antibody (Catalog # MAB6158) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. 3T3-L1 mouse embryonic fibroblast adipose-like cell line was stained with Mouse Anti-Human/Mouse UCP1 Monoclonal Antibody (Catalog # MAB6158, filled histogram) or isotype control antibody (Catalog # MAB0041, open histogram) followed by anti-Mouse IgG PE-conjugated Secondary Antibody (Catalog # F0102B). Expression of thermogenic markers was elevated following long-term rosiglitazone treatment and subsided upon beige (B) to white (W) transition. J774A.1 mouse reticulum cell sarcoma macrophage cell line was stained with Goat Anti-Mouse CD36/SR‑B3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2519, filled histogram) or isotype control antibody (Catalog #AB-108-C, open histogram), followed by Phycoerythrin-conjugated Anti-Goat IgG Secondary Antibody (Catalog # F0107).

CD36/SR‑B3 was detected in immersion fixed paraffin-embedded sections of mouse kidney using Goat Anti-Mouse CD36/SR‑B3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2519) at 3 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC004). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm and plasma membrane. Flow Cytometry: p53 Antibody (PAb 240) [NB200-103] - An intracellular stain was performed on HeLa cells with p53 Antibody [PAb 240] NB200-103 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-p53 (PAb 240) [NB200-103] at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

In the co-cultures, the expression of Ldha was reduced in cancer cells, but was elevated in fibroblasts. Bar = 50 um. Immunocytochemistry/Immunofluorescence: CD31/PECAM-1 Antibody (MEC 7.46) [NB100-1642] - MS1 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with at 5.0 ug/ml overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry-Frozen: CD31/PECAM-1 Antibody (MEC 7.46) [NB100-1642] - Frozen mouse lung tissues of WT B6 and IL1R--/- (knockout) mice were blocked with 1% BSA in PBS and were double stained with Alex647-conjugated CD31/PECAM-1 antibody (MEC 7.46) and PE-conjugated F4/80 for 2 hours at the room temperature. Image using the Alexa Fluor 647 format of this antibody.

Flow Cytometry: CD31/PECAM-1 Antibody (MEC 7.46) [NB100-1642] - A surface stain was performed on MS1 cells with CD31/PECAM-1 [MEC 7.46] Antibody NB100-1642AF647 (blue) and a matched isotype control (orange). Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

CD31/PECAM-1 (MEC 7.46) was detected in immersion fixed MS1 mouse pancreas/Islet of Langerhans endothelial cell line using Rat anti-CD31/PECAM-1 (MEC 7.46) Protein-G purified Monoclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NB100-1642AF647) (light blue) at 2 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Serum-starved MCF-7 human breast cancer cells were unstimulated (blue open histogram) or activated with 100 ng/mL Recombinant human IGF-1 (Catalog # 291-G1, dark orange filled histogram) for 15 minutes, then stained with Rabbit Anti-Human/Mouse/Rat Phospho-Akt (S473) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF887) followed by PE-conjugated Anti-Rabbit IgG Secondary Antibody (F0110). To facilitate intracellular staining, cells were fixed with formaldehyde (FC004) and permeabilized with Methanol.

Simple Western lane view shows lysates of MCF-7 human breast cancer cell line and A549 human lung carcinoma cell line untreated (-) or treated (+) with 100 ng/mL Recombinant Human IGF-I (291-G1) for 15 minutes, loaded at 0.2 mg/mL. A specific band was detected for Phospho-Akt (S473) at approximately 60 kDa (as indicated) using 5 µg/mL of Rabbit Anti-Human/Mouse/Rat Phospho-Akt (S473) Pan Specific Antigen Affinity-purified Polyclonal Antibody (Catalog # AF887). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

DDC up-regulates MMP-1 through ERK1/2 and Akt activation(A) Co-cultures were treated with 100 μM DDC for the indicated time.
