Application Notes
In Western Blot analysis, a band is recognized at ~68 kDa representing HIF-3 alpha. Because a band at ~71 kDa is also seen in hypoxic samples, it is important to run a negative control lysate as well (see image). In ICC/IF, cytoplasmic and nuclear staining was observed in HeLa cells, which is consistent with the literature under normoxic conditions for HIF-3 alpha (PMID: 16775626).

Background

Hypoxia-inducible factor (HIF) is one of the most important factors in the cellular response to hypoxia, transcriptionally activating genes encoding proteins that mediate adaptive responses to reduced oxygen availability. HIF is a heterodimer consisting of one of three subunits, HIF1 alpha, HIF2 alpha, or HIF3 alpha. HIF target genes play critical roles in metabolism, angiogenesis, cell proliferation and cell survival. HIF3 alpha protein is one of several alpha/beta-subunit heterodimeric transcription factors that regulate many adaptive responses to low oxygen tension (hypoxia). The alpha 3 subunit lacks the transactivation domain found in factors containing either the alpha 1 or alpha 2 subunits. HIF3 alpha may be a marker for tumor growth and angiogenesis.

  1. I'm testing HIF-3A antibody (NB100-2529). I have query about its specificity and expected size: mostly I see band at about 40 kDa instead of 71 (the samples are proteins from cell cultures grown in hypoxic conditions). In your control HIF-3A lysates one H00064344-T02 detecting band at 73 kDa and other H00064344-T01 at 37kDa. Since I'm running the SDS-PAGE, the band at 40kDa might be correct?
    • This protein has multiple isoforms (7) and they might have different expression patterns in various cell lines and tissue. As far as our overexpression control lysates are concerned, H00064344-T02 detects an isoform which is 72kd whereas in H00064344-T01 most probably isoform 4 which is around 40 KD is detected. It is also important to keep in mind that isoform 4 attenuates the binding of HIF1A to hypoxia-responsive elements (HRE), thus inhibiting HRE-driven transcription. Hypoxia induces down-regulation of isoform 4, leading to activation of HIF1A in hypoxia. Conversely, upon restoring normoxia, the expression of isoform 4 increases and thereby secures an inhibition of HIF1A activity. Please see this UniProt link for more information on HIF3A.
