![Western Blot: Calnexin Antibody - BSA Free [NB100-1974] - Immunoblot analysis of livers from control & L-Chrebp−/− mice subjected to fasting & refeeding with a high-sucrose diet. Littermate control & L-Chrebp−/− mice (same as those described in supplemental Tables S2A & S2B) were subjected to fasting & refeeding. The nonfasted (N) groups were fed chow diet ad libitum. The fasted (F) group was fasted 12 h, & the refed (R) group was fasted for 12 h & then refed with 60% (w/w) high-sucrose diet for 12 h prior to study. Liver whole-cell lysates & membrane fractions were prepared individually, & equal amounts of protein from each mouse of the same group (four per group) were pooled. Aliquots (40 μg for whole-cell lysates & 30 μg for membrane fractions) of the pooled protein were subjected to SDS-PAGE & immunoblot analysis. Immunoblot analysis of Insig-1 & Insig-2 were carried out using membrane fractions. Whole-cell lysates were used to detect other proteins. VMN neurons were identified in situ for glutamate decarboxylase65/67 (GAD65/67)- (top row, Panel 1A) or neuronal nitric oxide (nNOS)-immunoreactivity (-ir) (bottom row, Panel 2A); representative GAD65/67- or nNOS-ir-positive neurons are indicated by blue arrows. Areas shown in Panels 1A & 2A were rephotographed after positioning of a continuous laser track (depicted in green) around a single GAD65/67-ir (Panel 1B, blue arrow) or nNOS-ir neuron (Panel 2B, blue arrow) & subsequent ejection of the encircled cell by laser pulse (Panels 1C & 2C). Note that this microdissection technique causes negligible destruction of surrounding tissue & minimal inclusion of adjacent tissue. PVDF membrane was probed with 2.0 ug/mL of Mouse Anti-Human Notch-1 Monoclonal Antibody (Catalog # NB100-78486) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog #HAF018). Specific band was detected for Notch-1 at approximately 110 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. ![Immunohistochemistry-Paraffin: Notch-1 Antibody (mN1A) [NB100-78486] - Analysis of FFPE human pancreatic cancer using 1:10 dilution of Notch-1 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) was performed using Aperio AT2 (Leica Biosystems). Cytoplasmic staining in epithelial cells was observed. ![Immunocytochemistry/Immunofluorescence: TRPM2 Antibody [NB110-81601] - Neuro2a cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. The cells were incubated with anti-TRPM2 at 5 ug/mL overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue).
