![Immunocytochemistry/Immunofluorescence: Aromatase Antibody [NB100-1596] - U2OS cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-Aromatase Antibody NB100-1596 at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. ![Immunoprecipitation: LC3B Antibody (1251A) [NBP2-46892] - Western blot analysis of LC3 immunoprecipitation. 10 ug of monoclonal anti-LC3B antibody (clone 1251A) [Catalog # NBP2-46892] was used to immunoprecipitate LC3 from 200 ug of total protein from HeLa cells treated with or without 50 uM chloroquine for 24 h. Antibody-protein was captured by magnetic protein A/G beads, washed and subjected to SDS-PAGE on a 4-15% gel. Protein was transferred to 0.2 um PVDF membrane and probed with 2 ug/ml anti-LC3B (NB100-2220) and detected with an anti-rabbit HRP conjugated secondary antibody using chemiluminescence. Adult kidney sections were stained for calbindin‐D28K, Atp6v1b1, Aqp2, & Myh14 to identify intercalated cells within the distal & connecting tubule segments as well as to confirm Myh14 localization to the intercalated cell type. Cells with positive staining for Atp6v1b1 (red) can be found in tubule segments positive for calbindin‐D28K only (A–D) as well as in tubules positive for both calbindin‐D28K & Aqp2 (E–H); in both cases Atp6v1b1 staining is observed along the basolateral membrane indicating beta ‐ intercalated cell type (arrowhead, D&H). We also observed Atp6v1b1 staining in majority of the tubules along the apical membrane indicating the alpha ‐ intercalated cell type (arrowhead, I–L). Myh14 localized to the apical membrane of the intercalated cell type in the distal tubule (arrowhead, M–P). VMN neurons were identified in situ for glutamate decarboxylase65/67 (GAD65/67)- (top row, Panel 1A) or neuronal nitric oxide (nNOS)-immunoreactivity (-ir) (bottom row, Panel 2A); representative GAD65/67- or nNOS-ir-positive neurons are indicated by blue arrows. Areas shown in Panels 1A & 2A were rephotographed after positioning of a continuous laser track (depicted in green) around a single GAD65/67-ir (Panel 1B, blue arrow) or nNOS-ir neuron (Panel 2B, blue arrow) & subsequent ejection of the encircled cell by laser pulse (Panels 1C & 2C). Note that this microdissection technique causes negligible destruction of surrounding tissue & minimal inclusion of adjacent tissue. Western blot analysis of triplicate cell lysate pools produced from immunolabeled GAD65/67 (Panel 1D) or nNOS (Panel 2D) neurons from Veh/Veh animals showed that these proteins are expressed in VMN neuron samples obtained by combinatory immunocytochemistry/laser-catapult microdissection. ![Immunohistochemistry-Paraffin: TDP-43/TARDBP Antibody [NB110-55376] - Analysis of FFPE human placenta using TDP-43 antibody at 1:250 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Nuclear staining was observed.
