SYCP3 is a protein essential for synapsis of homologous chromosomes & gamma H2AX localizes to double-strand breaks & XY bodies during meiosis. A total of 200 spreads were counted in testis cell preparations from WT & GCKO mice at P22. (B) Chi square analysis shows GCKO testes contained significantly higher numbers of germ cell spreads at the leptotene & zygotene stages of meiosis I & fewer spreads at pachytene, diplotene & metaphase I stages (P<0.001), suggesting that the loss of Dicer1 lead to disruptions in progression through meiosis I. CXCR4 protein expression levels in lung tissue were examined by western blot. GAPDH served as an internal control (c, d; n = 3). Data are presented as the mean ± SD. #Value significantly greater than the corresponding value in saline-treated normoxic mice (##P 50 CTCs/ 2.5 ml blood) was processed via ficoll density centrifugation & the PBMC fraction was collected. Immunomagnetic anti-CD45 depletion was performed on the obtained PBMCs & the remaining cells were washed, cytospunned onto the slides. Slides were stained for PSMA, EpCAM, sLex, & CXCR4 using the protocol as described in Figure S1. MDA, PC3, & KG1 cells were simultaneously stained as a control for the following markers: PSMA= Magenta, EpCAM= Yellow, HECA-452= Green, CXCR4= Red. ![Immunocytochemistry/Immunofluorescence: Nestin Antibody (4D11) [NBP1-92717] - Mixed cultures of neonatal rat neurons and glia stained with NBP1-92717 (red), chicken antibody to Vimentin (NB300-223, green) and DNA (DAPI stain, blue). Astrocytes and neuronal stem cells stain strongly and specifically in a clearly filamentous fashion with the Nestin (4D11) antibody. The filamentous staining pattern is as expected as both Nestin and Vimentin are components of 10nm filaments. Note that some cells contain Nestin, but do not stain strongly for Vimentin and so appear red. Others contain Vimentin and not Nestin and so appear green - these are likely to be fibroblastic or endothelial cells. These neurons are capable of regenerating their axons and also, concomitant with regeneration, strongly upregulate alpha-Internexin in their perikarya. ![Immunocytochemistry/ Immunofluorescence: Calbindin D-28K Antibody [NBP2-50028] - Myh14 localizes to apical membranes of Atp6v1b1‐positive intercalated cells in the distal tubule. Adult kidney sections were stained for calbindin‐D28K, Atp6v1b1, Aqp2, & Myh14 to identify intercalated cells within the distal & connecting tubule segments as well as to confirm Myh14 localization to the intercalated cell type. Cells with positive staining for Atp6v1b1 (red) can be found in tubule segments positive for calbindin‐D28K only (A–D) as well as in tubules positive for both calbindin‐D28K & Aqp2 (E–H); in both cases Atp6v1b1 staining is observed along the basolateral membrane indicating beta ‐ intercalated cell type (arrowhead, D&H). We also observed Atp6v1b1 staining in majority of the tubules along the apical membrane indicating the alpha ‐ intercalated cell type (arrowhead, I–L). Myh14 localized to the apical membrane of the intercalated cell type in the distal tubule (arrowhead, M–P). Sections were mounted with vectashield containing DAPI to stain the nucleus (blue; D, H, L & P). Scale bar = 10 μm. Adult kidney sections were stained for calbindin‐D28K, Aqp2 & one of the NM2 isoforms. Myh9 (A–D) was not expressed in tubules positive for calbindin‐D28K & Aqp2; although adjacent tubules were positive for Myh9 (arrow). Myh10 (E–H) was expressed at low levels at the cell–cell junctions in calbindin‐D28K only positive tubules (white arrowhead), & in tubules positive for both calbindin‐D28K & Aqp2, Myh10 localized to cell–cell junctions, apical, & basolateral membrane (F & H, yellow arrowhead). Myh14 (I–L) was expressed on the apical membrane of calbindin‐D28K & Aqp2 null cells (arrowheads) in the distal tubular segments.
