![Western Blot: LC3A Antibody - BSA Free [NB100-2331] - LC3A Antibody [NB100-2331] - High autophagosome concentration is consumed during early immortalized human mesenchymal stem cell differentiation. Immortalized human mesenchymal stem cells were differentiated under osteogenic conditions (see Materials and methods) and assayed for changes in LC3I and LC3II during a 72-hour window. Cells were differentiated under standard conditions (top) or with addition of 5 uM rapamycin (middle) or 5 nM bafilomycin (bottom) for the first 3 hours of differentiation to modulate autophagy. Immunoblots were performed for LC3 at the indicated time points to assess autophagosome degradation via relative changes in LC3II (lower band; 17 kDa). Studies were repeated three times with similar trends seen consistently. ![Immunocytochemistry/ Immunofluorescence: GFAP Antibody [NB300-141] - Increased astrocyte activation in IL-1 beta -stimulated two-week old offspring is reduced by maternal LB supplementation. Representative images of fluorescence microscopy of claudin-5+ (location of the brain capillaries, red), GFAP+ astrocyte (green), & DAPI (nuclei, blue). Three to five sections per mouse were examined & at least three mice were examined in each group. Stronger than control SPF GFAP staining was observed around the blood vessel (see arrow) after IL-1 beta insult. Maternal supplemented group (LB) with or without postnatal insult had GFAP levels similar to the control group indicating that LB supplementation prevented astrocyte activation around the BBB endothelium. The membranes were blotted with NB100-56724 (lanes 1 & 2) or NB100-56507 (that recognizes both non-phospho and phosphorylated forms of IkBa) and immunoreactivity was detected by ECL. Following antigen retrieval, tissues blocked in 3% BSA in PBST for 30 mins at RT & then probed with Heat Shock Protein 86 (Hsp86) polyclonal antibody at 1:100 for 1 hr in a humidified chamber (right panel). As a -ve control, primary antibody eliminated from the staining procedure (left panel). Tissues washed with PBS/0.025% Tween-20 & endogenous peroxidase activity quenched with Peroxidase Suppressor for 30 mins at RT. Detection performed using an HRP-conjugated goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:250 followed by colorimetric detection using Metal Enhanced DAB Substrate Kit. Tissues were counterstained with hematoxylin & prepped for mouting. ![Immunoprecipitation: HSP90 alpha Antibody [NB120-2928] - Analysis of Heat Shock Protein (HSP86) was performed on HeLa cells. Antigen-antibody complexes formed by incubating 500ug whole cell lysate with 2ug of HSP86 polyclonal antibody overnight on a rocking platform at 4C. The immune complexes were captured on 50ul Protein A/G Plus Agarose, washed extensively, and eluted with Lane Marker Reducing Sample Buffer. Samples were then resolved on a 4-20% Tris-HCl polyacrylamide gel, transferred to a PVDF membrane and blocked with 5% BSA/TBST for at least 1 hour. The membrane was probed with a HSP86 polyclonal antibody at a dilution of 1:1000 overnight rotating at 4C. The membrane was washed in TBST, and probed with Clean-Blot IP detection Reagent at a dilution of 1:1000 for at least 1 hour.
