![Immunocytochemistry/Immunofluorescence: Fibronectin Antibody (2F4) [NBP2-22113] - Analysis of cultured HepG2 cells using Fibronectin antibody (clone 2F4) at 1:100 dilution. The cells were fixed in formalin and permeabilized using PBS with 0.1% Triton X-100 before incubating them with Fibronectin antibody or the mouse IgG1 isotype control antibody. DyLight 488 conjugated goat anti-mouse IgG (H+L) cross-adsorbed secondary antibody was used at 1: 200 dilution to detect the antigen-primary antibody complexes and the cells were counterstained with DAPI for labeling the nuclei. ![Immunoprecipitation: Pax6 Antibody [NB300-750] - Analysis of PAX6 was performed on 293T cells. Antigen: antibody complexes were formed by incubating 500ug whole cell lysate with 5ug of PAX6 polyclonal antibody overnight on a rocking platform at 4C. The immune complexes were captured on 50ul Protein A/G Plus Agarose, washed extensively, and eluted with 5X Lane Marker Reducing Sample Buffer. Samples were resolved on a 4-20% Tris-HCl polyacrylamide gel, transferred to a PVDF membrane, and blocked with 5% BSA/TBS-0.1%Tween for at least 1 hour. ![<b>Genetic Strategies Validation. </b>Immunohistochemistry-Paraffin: Noxa Antibody [NB100-56368] - IHC analysis of formalin fixed paraffin-embedded (FFPE) human tonsil using Noxa antibody at 1:50 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10 mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems).
