Immunocytochemistry/Immunofluorescence: O-GlcNAc Antibody (RL2) [NB300-524] - Neuro2a cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton X-100. The cells were incubated with anti-O-GlcNAc (RL2) at 5 ug/mL overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow Cytometry: O-GlcNAc Antibody (RL2) [NB300-524] - An intracellular stain was performed on Neuro2a cells with O-GlcNAc Antibody [RL2] NB300-524 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).

Western Blot: O-GlcNAc Antibody (RL2) [NB300-524] - Analysis of mouse cortical brain lysates using O-Linked N-Acetylglucosamine Monoclonal Antibody. Blots containing cortical extracts from 4 individual C57BL/6 mice (Lanes 1-4) were blocked with 5% milk in TBST, and probed with MA1-072 at 1:1000, followed by a fluorophore-conjugated goat anti-mouse IgG secondary antibody. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. 1 ug of antibody was added to 100 uL of staining buffer and cells were incubated for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Immunocytochemistry/Immunofluorescence: TGN46 Antibody [NBP1-49643] - U-2 OS cells were transfected with siRNAs targeting UGCG (siUGCG) or a non-targeting control (siNegCtrl) and plated onto glass coverslips. At 72 hours post-transfection cells were chilled to 4 degrees C on ice and rVSV-SFTSV was bound by centrifugation (1200xg, 30', 4 degrees C). Following centrifugation, media was replaced with pre-warmed media (37 degrees C) and the cells placed in a 37 degrees C incubator for 40 minutes before fixation in 1% paraformaldehyde for 15 minutes. Cells were then immunostained for viral antigen (anti-VSV M, red), TGN46 (green), and nuclei stained with DAPI (blue). Images are representative from at least 3 independent experiments. The cells were incubated with TGN46 Antibody conjugated to Alexa Fluor 488 (NBP1-49643AF488) at 2 ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow (Intracellular): TGN46 Antibody [NBP1-49643] - An intracellular stain was performed on U-937 cells with NBP1-49643 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by rabbit IgG APC-conjugated secondary antibody (F0111, R&D Systems).

Flow (Intracellular): TGN46 Antibody [NBP1-49643] - An intracellular stain was performed on HepG2 cells with TGN46 Antibody NBP1-49643AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

TGN46 was detected in immersion fixed Caco-2 human colorectal adenocarcinoma cell line using Rabbit anti-TGN46 Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 488 (Catalog # NBP1-49643AF488) (green) at 2 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

TGN46 was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rabbit anti-TGN46 Affinity Purified Polyclonal Antibody conjugated to Biotin (Catalog # NBP1-49643B) at 2 µg/mL overnight at 4C. Cells were stained using Streptavidin conjugated to DyLight 550 (red) and counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunofluorescence microscopy of incoming virus particles, 20 minutes.(A, B) A549 cells were plated onto glass coverslips and the following day replaced with media containing NB-DNJ (200μM) or left untreated. Forty-eight hours later, cells were chilled to 4C on ice, then rVSV-SFTSV was bound by centrifugation (1200xg, 30’, 4C). Following centrifugation, media was replaced with pre-warmed media (37C) and the cells placed in a 37C incubator for 20 minutes before fixation in 2% paraformaldehyde for 10 minutes. Immunofluorescence microscopy of rVSV-SFTSV particles following UGCG knockdown.(A-D) U-2 OS cells were transfected with siRNAs targeting UGCG (siUGCG) or a non-targeting control (siNegCtrl) and plated onto glass coverslips. CulD, N-terminal-fused w/ the LexA activation domain, expressed under control of the constitutive ADH promoter. A. nidulans proteins expressed in S. cerevisiae wild type & delta csn5 background. Western analysis w/ antibodies against Rub1 ( alpha -Rub1), the LexA epitope ( alpha -LexA) & the V5 epitope ( alpha -V5) performed. Detection w/ alpha -Rub1 generated two additional signals upon culD expression, representing LexA-CulD & a second CulD pool where LexA unspecifically cleaved off. Both signals disappeared upon co-expression of DenA indicating deneddylation activity (red arrows). The slower migrating signal of alpha -LexA western experiments corresponded to Rub1 modified LexA-CulD. This signal absent when DenA co-expressed. Y27632 (Y10; 10 μM) & calyculin A (A50; 50 μg/ml) were used as negative & positive controls. Western blot analysis & staining with anti-phospho-MLC, anti-total-MLC, & anti-beta actin antibodies was then performed. Phospho-MLC, total-MLC, & beta actin were all obtained with their appropriate protein size bands. The relative Phospho-MLC intensity (%) was expressed as [(Phospho-MLC/total-MLC)drug treated/ (Phospho-MLC/total-MLC)cell only x 100%]. Cells were broken by glass beads treatment & analyzed by 32P-ADP-ribosylation in the presence of additionally added purified wild type yeast actin (1 μg). Labeled bands represent modified yeast actin & confirm intracellular production of functionally active Ia by the S. cerevisiae strains. (B) Production of Ia by the wild type S. cerevisiae strain. Wild type yeast strains harboring the Ia-containing plasmid (Ia) or the control vector (vector) were cultivated in glucose-containing liquid medium until OD595 = 0.5. <b>Genetic Strategies Validation. </b>Simple Western: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - Detection of Ki67/MKI67 by Simple WesternTM. Simple Western lane view shows lysates of HeLa parental cell line and Ki67 knockout (KO) HeLa cell line. A specific band was detected for Ki67/MKI67 at approximately 317 kDa (as indicated) in the parental cell line, but is not detectable in the knockout HeLa cell line using 1:100 of Mouse Anti-Ki67/MKI67 Monoclonal Antibody (Catalog # NBP2-22112). GAPDH is shown as a loading control. This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Immunocytochemistry/Immunofluorescence: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - NIH3T3 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti- NBP2-22112 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/Immunofluorescence: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - A431 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti- NBP2-22112 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

The cells were incubated with anti-Ki-67 (8D5) conjugated to Alexa Fluor 488 [NBP2-22112AF488] at 10ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - Ki67 antibody was tested at 1:100 in HeLa cells with DyLight 488 (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and DyLight 550 (red). Image objective 40x.

Ki67/MKI67 (8D5) was detected in immersion fixed A431 human skin carcinoma cell line using Mouse anti-Ki67/MKI67 (8D5) Protein-G purified Monoclonal Antibody conjugated to Alexa Fluor® 488 (Catalog # NBP2-22112AF488) (green) at 5 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

RPTECs trigger CD4+ T-cells to proliferate, anoxia-reoxygenation increases proliferation further, whereas CD4+ T-cell differentiation to regulatory T-cells does not occur. CD4+ T-cells were cultured alone or co-cultured with RPTECs for 7 days. RPTECs were previously subjected or not to 90 min of anoxia. Four such experiments were performed. Compared to isolated CD4+ T-cell culture, proliferation index was higher in RPTEC CD4+ T-cell co-culture and increased further in the case of previous RPTEC culture under anoxia. * indicates a p < 0.05 compared to isolated CD4+ T-cell culture, # a p < 0.05 compared to RPTEC CD4+ T-cell co-culture, and ^ a p < 0.05 compared to RPTEC (A). Previous culture of RPTECs under anoxia induced further Ki-67 in CD4+ T-cells (B,C). FOXP3 expression in CD4+ T-cells remained unaffected under all conditions (B,D). The Western blotting results were normalized for beta -actin and then depicted after normalization for the control group. Albumin overload induces cellular senescence and EMT. Panel (A) depicts the results of one out of three performed experiments. Anoxia-reoxygenation induces senescence phenotype, while LW6 prevents it. Flow (Intracellular): alpha Tubulin Antibody (YOL1/34) [NB100-1639] - An intracellular stain was performed on HeLa cells with Tubulin [YOL1/34] Antibody NB100-1639 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Rat IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 488.

Flow Cytometry: alpha Tubulin Antibody (YOL1/34) [NB100-1639] - An intracellular stain was performed on HeLa cells with alpha Tubulin Antibody [YOL1/34] NB100-1639AF647 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Cells are assayed for spindle morphology and Cdc14 release phenotype by indirect immunofluorescence with anti-tubulin or anti-HA, which recognizes the HA-tagged version of Cdc14 in all strains. Cells are visualized by nuclear staining with DAPI and by differential interference contrast (DIC) optics for comparison. No release refers to dense nucleolar staining of Cdc14-HA, full release refers to cells with uniform Cdc14-HA localization throughout the nucleus, while partial release refers to cells with both nuclear and residual nucleolar staining. Sfu1 and Ssn3 mediate opposing effects on Sef1 stability and virulence.a) Sef1 protein is stabilized in an sfu1 delta delta mutant. Wild-type or sfu1 delta delta strains containing Sef1-Myc were propagated in iron-replete medium containing 2 mg/ml cycloheximide. Samples were recovered at the indicated time points, and Sef1-Myc was visualized using monoclonal antibodies against the Myc epitope, followed by incubation with secondary antibodies that were coupled to infrared dyes and quantified using a Li-Cor instrument. Note that a single band corresponding to higher mobility (unphosphorylated) Sef1 is present in both strains. b) Sef1 protein is destabilized in the ssn3 delta delta mutant. The experiment was performed as above, except that cells were propagated in iron-depleted medium containing cycloheximide. Note that phosphorylated Sef1 recovered from wild-type cells runs with slower mobility. c) Model for Sef1 regulation by Sfu1 and Ssn3 under iron-replete vs. iron-depleted conditions. Immunocytochemistry/ Immunofluorescence: SCP3/SYCP3 Antibody [NB300-231] - Meiotic spreads show deletion of Dicer1 disrupts progression of meiosis I.(A) It was possible to identify the 5 sub-stages of meiosis I by combining the reactivity patterns of two antibodies against synaptonemal complex protein 3 (SYCP3) & gamma H2AX. SYCP3 is a protein essential for synapsis of homologous chromosomes & gamma H2AX localizes to double-strand breaks & XY bodies during meiosis. A total of 200 spreads were counted in testis cell preparations from WT & GCKO mice at P22. (B) Chi square analysis shows GCKO testes contained significantly higher numbers of germ cell spreads at the leptotene & zygotene stages of meiosis I & fewer spreads at pachytene, diplotene & metaphase I stages (P<0.001), suggesting that the loss of Dicer1 lead to disruptions in progression through meiosis I.
