Application Notes
WB:Approx 90 kDa band observed in Human Brain (cerebellum), Heart and Rat Brain lysates (calculated MW of 91.7 kDa according to NP_060676.2).
IHC: In paraffin embedded Human Testis, shows positive staining in Leydig cells, and cytoplasmic staining (dot-like) of some epithelial cells of the epididymis and of the rete testis. Data obtained with a previous batch.
Immunocytochemistry: Vesicular staining of VPS35 (green) in HEK293 and in SHSY5Y. Based on these data, this antibody was endorsed by PabmAbs (http://pabmabs.com/wordpress/?p=1523).

Background

Chemokine receptor D6 expression has been documented in human skin, small intestine, large intestine, appendix, lymph node, vessels, tonsil, spleen, as well as from rat brain tissues. ESTs have been isolated from cancerous human lung, uterus, cervix, placenta, blood, and from normal human placenta libraries. Caution: G protein-coupled receptor D6 was previously called CCR10 and/or CCR9. CCR9 and C-C chemokine receptor 9 referred to both chemokine receptor D6 and the unrelated receptor GPR-9-6.

VPS35 antibody (Imgenex: IMG-3575) was used for ICC-IF analysis of Huh 7.5.1 cells, JFH1-plasmid infected Huh 7.5.1 cells, OR6 cells (which harbors the full-length genotype 1b HCV RNA) and Cured OR6 cells (OR6 cells treated with 10 IU/ml of IFN alpha for 7 days) [Fig 2A/B, 3A/B].

VPS35 antibody was used for ICC-IF on HeLa cells that were transfected with a plasmid expressing mock/GFP or GFP-VPS26. The antibody was also used for WB application on lysates of HeLa cells which were either mock transfected /Control or transfected with siRNA duplexes targeting CypB (Si-CypB), SNX1 (Si-SNX1), WASH (Si-WASH), or Vps35 (Si-Vps35) [FIG 6].
