Background
Zinc finger protein 521 (ZNF521; EHZF) is an 1131 amino-acid long transcription factor that includes 30 zinc fingers which are scattered along its sequence. Zfp521 appears to play a role in development, bone formation, erythroid cell differentiation, stem cell regulation and oncogenesis. Zfp521 is expressed in developing bones, where it binds to Runx2, thusly repressing the transcriptional activity of Runx2. This balance between Zfp521 and Runx2 may play a role in the regulation of osteoblast differentiation and bone formation (PMID: 20392215). Zinc finger protein 521 has also been shown to be expressed in hematopoietic stem and progenitor cells but is down-regulated as they go through the process of differentiation (PMID:19342626). Furthermore, ZNF521 was shown to inhibit the activity of GATA-1 which is a crucial regulator of erythroid differentiation by directly binding to GATA-1 (PMID: 19049973). Expression of ZNF521 has been detected in most acute myelogenous leukaemias, as well as in medulloblastomas and other brain tumors (PMID: 17543573).
