Notes
HEK293T cells in 10-cm dishes were transiently transfected with a non-lipid polymer transfection reagent specially designed and manufactured for large volume DNA transfection. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix, 1mM PMSF and 1mM Na3VO4, and then centrifuged to clarify the lysate. Protein concentration was measured by BCA protein assay kit.This product is manufactured by and sold under license from OriGene Technologies and its use is limited solely for research purposes.

Background
Mitochondrial uncoupling proteins (UCP) are members of the larger family of mitochondrial anion carrier proteins (MACP). UCPs separate oxidative phosphorylation from ATP synthesis with energy dissipated as heat, also referred to as the mitochondrial proton leak. UCPs facilitate the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. They also reduce the mitochondrial membrane potential in mammalian cells. Tissue specificity occurs for the different UCPs and the exact methods of how UCPs transfer H+/OH- are not known. UCPs contain the three homologous protein domains of MACPs. This gene is expressed in many tissues, with the greatest expression in skeletal muscle. It is thought to play a role in nonshivering thermogenesis, obesity and diabetes. Chromosomal order is 5'-UCP3-UCP2-3'. [provided by RefSeq]. Hi, I'd like to measure UCP2 levels in various mouse tissues by western blot. I was wondering which of your antibodies would be best for this purpose. In the literature I have seen some papers where they have isolated mitochondria to be able to see UCP2 and others where they have not. We would like to use frozen tissue samples, so isolating mitochondria will not be an option. We would like to use liver, muscle, pancreas, kidney, brown fat, subcutaneous fat, and gonadal fat. I'd appreciate any advice you might have.
- Regarding your question on our UCP2 antibodies, we do have a product (catalog # NB100-78377), but our lab confirmed that it has only been validated in mouse liver and brain and there is no guarantee on other tissues.
