Background

RANK Ligand (RANK L), also known as TRANCE, TNFSF11, OPGL, and ODF, is an approximately 40 kDa transmembrane protein in the TNF cytokine superfamily. RANK L associates into trimers, and multiple soluble forms can be generated by ADAM10, MMP14, or MT1-MMP mediated cleavage. Both transmembrane and soluble RANK L act through RANK receptors and are antagonized by the decoy receptor, Osteoprotegrin. Within the ECD, human and mouse RANK L share 85% aa sequence identity. RANK L is produced by osteoblasts, bone marrow stromal cells, activated T cells, mammary epithelial cells, and astrocytes. It is promotes osteoclast differentiation, dendritic cell activation, mammary gland function, and body temperature regulation.
