Application Notes
This TET1 antibody is useful for Immunohistochemistry on paraffin embedded sections where staining is observed in the nucleus and cytoplasm of mouse spleen tissue. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended. This antibody is not applicable for ICC/IF or Western Blot.

Background

TET1 (Tet methylcytosine dioxygenase 1, LCX, CXXC6) is a member of a novel protein family. This family of proteins may play a role in epigenetic regulation via modification of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (hmC). TET1 is a fusion partner of the MLL gene in acute myeloid leukemia (PMID: 19372391). It has been shown that TET1 supports DNA demethylation in mammalian cells via the base excision repair pathway where TET1 converts 5mC to hmC. Additionally, TET1 is integral to neuronal activity-induced, region specific, active DNA demethylation and subsequent gene expression in the dentate gyrus of the adult mouse brain in vivo. This suggests that the mechanism for active DNA demethylation in mammals is through TET1-induced oxidation-deamination (PMID: 21496894).
