T-cell leukemia/Lymphoma Protein 1A (TCL1A), also known as p14TCL1 is a product of the TCL1 gene that is involved in T-cell prolymphocytic leukemia (TPLL). T-PLL is a rare form of mature T-cell leukemia, which is consistently associated with chromosomal rearrangements characterized by the juxtaposition of the TCRA locus on chromosome 14q11 and the TCL1A gene on 14q32.13. TCL1 is overexpressed in Burkitt lymphoma, the majority of AIDS-related non-Hodgkin lymphoma-designated immunoblastic plasmacytoid lymphoma, lymphoblastic lymphoma, chronic lymphocytic leukemia, mantle cell lymphoma, follicular lymphoma, diffuse large B-cell lymphoma, and primary cutaneous B-cell lymphoma.

Immunohistochemistry-Paraffin: AKT1 [p Ser473] Antibody (104A282) [NB100-56749] - IHC analysis of an FFPE human breast carcinoma tissue section using 1:250 dilution of phospho Ser473 AKT1 antibody (clone 104A282) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10 mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of phosphor-AKT (Ser-473) in the cancer cells as well as the stromal cells. Staining was performed by Histowiz.

Eight days after MPP+ administration, our Western blot results (MAB7566) show that MPP+ treatment reduced tyrosine hydroxylase and Bcl-2 expression in the ipsilateral side of the rat substantia nigra (SN), but not in the contralateral side. EA stimulation (50 Hz) enhanced mature BDNF, tyrosine hydroxylase, and Bcl-2 expression in the MPP+-treated ipsilateral side. Specific band was detected for c-Fox at approximately 52 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

The single band at 37 kDa represents GAPDH protein.

DAPI reveals nuclei of neurons and glia in blue. The hippocampal neurons stain green for FOX3/NeuN and a few also are expressing c-FOS, and so appear orange. These cells were spontaneously active at the time the animal was sacrificed.
