Immunocytochemistry/Immunofluorescence: Src Antibody (21H5) [NBP3-26303] - Staining of MCF-7 cells with Src Antibody (21H5) at 1:39, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 488-conjugated Goat Anti-Rabbit IgG (H+L).

Flow Cytometry: Src Antibody (21H5) [NBP3-26303] - Overlay histogram showing SH-SY5Y cells stained with Src Antibody (21H5) (red line) at 1:50. The cells were fixed with 70% Ethylalcohol (18h) and then permeabilized with 0.3% Triton X-100 for 2 min. The cells were then incubated in 1x PBS /10% normal goat serum to block non-specific protein-protein interactions followed by primary antibody for 1 h at 4C. The secondary antibody used was FITC goat anti-rabbit IgG (H+L) at 1/200 dilution for 1 h at 4C. Control antibody (green line) was used under the same conditions. Acquisition of >10,000 events was performed.

Background

Src is a protein tyrosine kinase known to regulate cellular adhesion. Composed of three major domains; SH2, SH3 and a kinase catalytic domain, Src can be switched from an inactive to an active state though control of its phosphorylation state or through protein-protein interaction. Phosphorylation of Y529 by Csk and Chk inactivates Src, while dephosphorylation of Y529 will modify its conformation to an open active state (1-3). Mutation of Y529 leads to Scr overactivity. Several cancers including colon and breast cancer have been associated with an increase of Src activity (4).
