Background

SPARC, also called Osteonectin, is an extracellular matrix (ECM) associated glycoprotein. SPARC activates matrix metalloproteinase-2, a key effector of ECM proteolysis, a process linked to enhanced tumor invasion and metastatic potential (1). MMP expression by tumor cells themselves may be representative of more invasive migratory cancers or latter stages of tumor progression (2, 3). SPARC appears to enhance MMP-2 activation at least in invasive breast cancer cell lines. Levels of SPARC expression appear to be directly correlated with progressive cancers not only of breast, but colon, kidney, ovary and brain (4, 5). SPARC specific antibody may establish levels of expression of SPARC providing its' use as a biomarker of late stage or aggressive tumors.
