Background

RICTOR is a component of a protein complex [mTORC2] that integrates nutrient and growth factor-derived signals to regulate cell growth. Rictor is a key component of mTORC2 which plays an essential role in Akt phosphorylation and signaling. Deregulation of Akt/protein kinase B (PKB) is implicated in the pathogenesis of cancer and diabetes. Rictor-mTOR may serve as a drug target in tumors that have lost the expression of PTEN, a tumor suppressor that opposes Akt/PKB activation.

• Comments: Mouse white adipose tissue was homogenised and protein content was quantified by a BCA assay. Twenty micrograms of protein were resolved on a 4-12% Bis-Tris gel and transferred to nitrocellulose membranes. Membranes were probed with primary antibody Rictor diluted 1:1000 in 5% BSA 2, before incubation with anti rabbit secondary horseradish peroxidase-conjugated antibody 1:5000. Blots were visualised with Immobilon Western Chemiluminescence HRP Substrate and imaged with Syngene chemiluminescence imaging system.
