| Application Notes |
A strong reaction is seen in sandwich ELISA in combination with a biotinlabeled antibody against protein S. Reactivity is Ca++ dependent and is optimal when using a tris-buffer containing 1 mM CaCl2. In Dot blotting SDS-treatment abolishes the reactivity |

75 kDa. Disclaimer note: The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors. |

Background
Protein S is a vitamin K-dependent, potent neutral anticoagulant plasma glycoprotein. Protein S has a molecular weight of approximately 70 kDa and consists of a single polypeptide chain. The concentration of protein S in human plasma is 25 ug/ml, with a half-life of approximately two days. About 40% of protein S in plasma is in a free form, whereas 60% is complexed with C4b-binding Protein (C4BP). Only the free protein S functions as a cofactor to antigen presenting cells (APC) in the APC-dependent degradation of Factor Va and Factor VIIIa.

-
Our customer interested in Protein S Antibody (8D5) #NBP1-05150 and would like to know whether its immunogen or epitope includes aa. 42-87 (Gla domain) of protein S precursor? Could you tell us if it could be disclosed?
- The immunogen that was used to generate our Protein S antibody with catalogue number NBP1-05150 was the full length protein, which was isolated from human plasma. We do not epitope map our antibodies, and as such I am unable to tell you exactly where on the protein the antibody binds.
