Method
Lyophilized from a 0.2 um filtered solution in PBS.
Details of Functionality
Measured by the ability of the immobilized protein to support the adhesion of HEK293 human embryonic kidney cells. Recombinant Human (rh) Osteopontin/OPN immobilized at 1 ug/mL, 100 uL/well, will mediate the adhesion of added 293 cells >30% (1 x 105 cells/well). Recombinant Human Coagulation Factor II/Thrombin (Catalog # 1473-SE) proteolytic treatment of this rhOsteopontin/his can increase HEK293 cell adhesion by about 5-fold.

Reconstitution Instructions
Reconstitute with 0.1 mg/ml in sterilized PBS containing at least 0.1% human or BSA to desired concentration.

Notes

This lyophilized preparation is stable at 2-8 degrees C, but should be kept at -20 degrees C for long term storage, preferably desiccated. Upon reconstitution, the preparation is most stable at -20 to -80 degrees C, and can be stored for one week at 2-8 degrees C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20 degrees C to -80 degrees C. Avoid repeated freeze/thaw cycles.

Background

Osteopontin is the principal phosphorylated glycoprotein of bone and is expressed in a limited number of other tissues including dentine. Osteopontin is produced by osteoblasts under stimulation by calcitriol and binds tightly to hydroxyapatite. It is also involved in the anchoring of osteoclasts to the mineral of bone matrix via the vitronectin receptor, which has specificity for osteopontin. Osteopontin is overexpressed in a variety of cancers, including lung, breast, colorectal, stomach, ovarian, melanoma and mesothelioma.
