Lane 1: HeLa cell lysate Lane 2: A549 cell lysate Lane 3: HT-29 cell lysate Lane 4: MEF cell lysate Lane 5: Human lung tissue lysate Lane 6: Mouse lung tissue lysate Lysates/proteins at 20 ug/Lane. Predicted band size: 243 kDa Observed band size: 270 kDa Exposure time: 5 minutes; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32651) at 1/2,000 dilution was used in 5% NFDM/TBST at 4 overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background

Ninein encodes one of the proteins important for centrosomal function. This protein is important for positioning and anchoring the microtubules minus-ends in epithelial cells. Localization of this protein to the centrosome requires three leucine zippers in the central coiled coil domain. Multiple alternatively spliced transcript variants that encode different isoforms have been reported.
