Western Blot: NFATC2/NFAT1 Antibody (JA11-08) [NBP2-66974] - Analysis of NFATC2/NFAT1 on different lysates with Rabbit anti-NFATC2/NFAT1 antibody at 1/500 dilution. Lane 1: MCF-7 cell lysate Lane 2: Daudi cell lysate Lysates/proteins at 10 ug/Lane. Predicted band size: 100 kDa Observed band size: 130 kDa Exposure time: 2 minutes; 6% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/Immunofluorescence: NFATC2/NFAT1 Antibody (JA11-08) [NBP2-66974] - Staining NFAT1 in MCF-7 cells (red). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Flow Cytometry: NFATC2/NFAT1 Antibody (JA11-08) [NBP2-66974] - Analysis of Jurkat cells with NFAT1 antibody at 1/50 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black). Alexa Fluor 488-conjugated goat anti rabbit IgG was used as the secondary antibody.

Background

The nuclear factor of activated T-cells (NFAT) transcription complex is required for the expression of a group of proteins that collectively regulate the immune response. Four NFAT proteins, encoded on separate genes and expressed as several splice variants, have been described: NFAT1, NFAT2, NFAT3, and NFAT4. These proteins show a low level of sequence similarity with the Dorsal/Rel/NFkB family of transcription factors. Another NFAT-related protein termed NFAT5 differs from isoforms 1-4 in that it lacks many of the Fos/Jun contact sites observed in its predecessors and its subcellular localization is not calcineurin-dependent.
