Western Blot: Mucolipin 1 Antibody [NB110-82375] - Total protein from mouse brain was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-Mucolipin 1 (NB110-82375) in blocking buffer and detected with an anti-rabbit HRP secondary antibody using NovaLume chemiluminescence detection reagent (NPB2-61915).

Background

Mucolipin 1 (MCOLN1, mucolipidin or TRPML1), is a member of TRP ion channels family and unlike most TRPs, mucolipin 1 primarily resides in the membranes of endocytic pathway (late endosomes/lysosomes). Widely expressed as multipass membrane protein, mucolipin 1's high expression levels are found in brain, liver and kidney. Mucolipin 1 activation by phosphatidylinositol 3,5-bisphosphate [PtdIns(3,5)P(2)], which is predominantly found in endocytic pathway and mucolipin 1 regulates the fusion/fission of endocytic pathway vesicles, and/or some aspect of lysosomal ion homeostasis such as pH, iron, or zinc content. It is a cation channel, and besides controlling membrane trafficking of proteins and lipids, it plays a major role in Ca2+ transport regulating lysosomal exocytosis. MCOLN1 mutation mediated mucolipin 1 down-regulation results in rare lysosomal storage disease mucolipidosis type IV (MLIV) that is characterized by progressive neurodegeneration with early onset, mental retardation, corneal opacity, retinal degeneration, vacuolization of parietal cells, constitutive achlorhydria and an elevated gastrin levels.
