Specificity
NB100-64952 recognizes a polymorphic determinant on the murine MHC (major histocompatibility complex) class I molecule. In the mouse, the MHC class I molecule is expressed on the cell surface of all nucleated cells but expression levels may differ according to cell type. Clone ER-MP52 specifically recognizes mouse strains with the haplotypes H-2^b,w16 and H-2 ^d,p,q. Mouse strains with the haplotypes H-2 ^f,r,s,w17,w23,w27 show weak reactivity with this antibody.

Background

MHC Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. MHC class I antigens are heterodimers consisting of one alpha chain (44kDa) with beta 2 microglobulin (11.5 kDa). The antigen is expressed by all somatic cells at varying levels. MHC Class I molecules are expressed on most nucleated cells where they present endogenously synthesized antigenic peptides to CD8+ T lymphocytes, which are usually cytotoxic T cells. Fibroblasts or neurons however only show a low level of antigen.

The blue is DAPI staining of nuclear DNA. DCX antibody reveals strong cytoplasmic staining in a population of small developing neurons and their processes, while MAP2 antibody stains dendrites and perikarya of mature neurons. Therefore DCX antibody is an excellent marker of developing neuronal cells.
Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Rat brain neural cultures stained with NBP1-92684 (green), a chicken polyclonal antibody to GFAP (NB300-213, red) and DNA (blue). The NBP1-92684 antibody reveals strong cytoplasmic staining in a population of small developing neurons. These cells are often found in small clumps as in these cultures, as in this example. Note that they are not positive for MAP2, which is characteristic of more mature neurons, and that the mature neuron shown is negative for NBP1-92684. This Doublecortin antibody is therefore an excellent marker of developing neuronal cells.
