Western Blot: Klotho Antibody (JM93-76) [NBP2-75551] - Analysis of Klotho on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Immunocytochemistry/Immunofluorescence: CD34 Antibody (MEC 14.7) [NB600-1071] - CD34 antibody was tested in WEHI-3 cells with DyLight 488 (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and DyLight 550 (red).
Immunohistochemistry-Paraffin: CD34 Antibody (MEC 14.7) [NB600-1071] - Analysis of a FFPE tissue section of mouse small intestine using rat anti-mouse CD34 (clone MEC 14.7) at 1:100 dilution. The signal was developed using HRP-conjugated anti-rat secondary with DAB reagent which followed counterstaining of nuclei using hematoxylin. This antibody specifically labelled the endothelial cells in blood vessels located primarily in the sub-mucosa, and of that of the mucosa muscularis and the mucosal lacteal.
Flow Cytometry: CD34 Antibody (MEC 14.7) [NB600-1071] - An intracellular stain was performed on Raw264.7 cells with CD34 Antibody [MEC 14.7] NB600-1071 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rat IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 488 (SA5-10018, Thermo Fisher).

Western blot shows lysates of MO7e human megakaryocytic leukemic cell line, P815 mouse mastocytoma cell line, and MC/9‑2 mouse mast cell line. PVDF membrane was probed with 0.1 µg/mL of Goat Anti-Human/Mouse CD117/c-kit Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1356) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). Specific bands were detected for CD117/c-kit at approximately 135, 150 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Simple Western lane view shows lysates of MO7e human megakaryocytic leukemic cell line, P815 mouse mastocytoma cell line and MC/9-2 mouse mast cell line, loaded at 0.2 mg/mL. A specific band was detected for CD117/c-kit at approximately 150-165 kDa (as indicated) using 5 µg/mL of Goat Anti-Human/Mouse CD117/c-kit Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1356) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.        
CD117/c-kit was detected in immersion fixed frozen sections of mouse embryo using Goat Anti-Human/Mouse CD117/c-kit Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1356) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Immunohistochemistry: Insulin Antibody (E2-E3 (same as INS04)) [NBP2-34260] - Immunohistochemistry of pancreas shows relatively higher reactivity for insulin in PrP-/- relative to Tg40 PrP sections (panels 1 & 3). Iron overloading decreases insulin reactivity in Tg40 PrP but not in PrP-/- samples (panels 1 vs. 2 & 3 vs. 4). Reaction for glucagon is higher in iron overloaded Tg40 PrP relative to control samples (panels 5 & 6). The difference is minimal in PrP-/- samples (panels 7 & 8).
