Western Blot: Integrin alpha 3/CD49c Antibody (A7H2) [NBP3-32475] - Western blot analysis of Integrin alpha 3/CD49c on different lysates with Mouse anti-Integrin alpha 3/CD49c antibody (NBP3-32475) at 1/1,000 dilution. Lane 1: HeLa cell lysate (10 ug/Lane) Lane 2: HUVEC cell lysate (10 ug/Lane) Lane 3: A431 cell lysate (10 ug/Lane) Lane 4: Human kidney tissue lysate (20 ug/Lane) Predicted band size: 117 kDa Observed band size: 130 kDa Exposure time: 28 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32475) at 1/1,000 dilution was used in 5% NFDM/TBST at 4 overnight. Goat Anti-Mouse IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/ Immunofluorescence: Integrin alpha 3/CD49c Antibody (A7H2) [NBP3-32475] - Immunocytochemistry analysis of U-87 MG cells labeling Integrin alpha 3/CD49c with Mouse anti-Integrin alpha 3/CD49c/CD49c antibody (NBP3-32475) at 1/100 dilution. Cells were fixed in 100% precooled methanol for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Mouse anti-Integrin alpha 3/CD49c antibody (NBP3-32475) at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Mouse IgG H&L (iFluor™ 488) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI. beta Tubulin (red) was stained at 1/100 dilution overnight at +4℃. Goat Anti-Rabbit IgG H&L (iFluor™ 594) were used as the secondary antibody at 1/1,000 dilution.

Background

Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. Alpha chain 3 undergoes post-translational cleavage in the extracellular domain to yield disulfide-linked light and heavy chains that join with beta 1 to form an integrin that interacts with many extracellular-matrix proteins. The alpha 3 beta 1 integrin is known variously as: very late (activation) antigen 3 (VLA3), very common antigen 2 (VCA2), extracellular matrix receptor 1 (ECMR1), and galactoprotein b3 (GAPB3).
