Background

IMPDH2, inosine monophosphate dehydrogenase, mediates negative regulation of activity via the signaling pathway (1). IMPDH2 upregulates phosphatase activity of which then goes on to inactivate PI3-k. An important function of IMPDH2 is the regulation of guanosine nucleotide pathways. Through conversion of IMP- inosine monophosphate to XMP-xanthosine monophosphate de novo synthesis of guanosine nucleotides is controlled and in T and B lymphocytes IMPDH2 activity affects proliferation (2). IMPDH2 and IMPDH specific detection and control has made it a therapeutic target for immunosuppressive, anti-cancer, anti-parasitic, or anti-viral therapy. IMPDH inhibitors such as ribavarin have been shown to potentiate anti-viral activity through induction of IRFs, further enhancing the anti-viral effects of type 1 Interferons (3). For example, monitoring IMPDH or IMPDH2 activity as a biomarker for mycophenolic acid-induced immunosuppression in renal transplants is one such use of IMPDH or IMPDH2 specific probes (4).
