ICAT/CTNNBIP1 Antibody (536017) [Unconjugated]

ICAT was detected in immersion fixed paraffin-embedded sections of human fibrocystic breast tissue using Mouse Anti-Human ICAT Monoclonal Antibody (Catalog # MAB3016) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm. Background

Human ICAT is a physiological inhibitor of the beta -Catenin/Wnt signaling pathway. It is 9 kDa in size and found in both cytosol and nucleus. The molecule contains an alpha ‑helical N-terminal region (aa 9‑54) and an acidic, extended C-terminal region (aa 55‑78). The alpha -helical region binds to beta -Catenin, displacing CBP/p300, while the acidic C-terminus interacts with the beta -Catenin N-terminus, blocking Tcf interaction. Human ICAT shares 97% aa sequence identity with mouse and bovine ICAT, respectively.
