Application Notes
Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For immunofluorescent staining, the suggested use of this reagent is: 1.0 microgram per one million cells in 100 microliter volume or 100 microliter of whole blood. Immunohistochemistry-frozen/paraffin. It is recommended that the reagent be titrated for optimal performance for each application.

Background

The HI43 monoclonal antibody recognizes human HLA-DP also known as MHC class II DP monomorphic antigen. The major histocompatability complex is composed of two heterodimeric glycoproteins ( alpha and beta chains) with apparent molecular weights of 28 and 34 kD. HLA-DP is expressed on hematopoietic progenitors, B cells, monocytes and macrophages, dendritic cells, Langerhans cells, activated T cells, and some endothelial and epithelial cells. HLA-DP is not expressed on resting T cells, erythrocytes and platelets. HLA-DP is highly expressed on some B cell leukemias and some chronic myelogenous leukemias, may be useful in the diagnosis and classification of leukemias. HLA-DP presents peptide fragments mainly from degraded intravesicular and extracellular proteins to CD4+ lymphocytes. Specific HLA-DP alleles have implicated in the pathogenesis of several autoimmune diseases including juvenile rheumatoid arthritis and chronic immune disorders such as berylliosis. The HI43 antibody has been reported to be useful for flow cytometric detection of human HLA-DP and immunohistochemistry (acetone-fixed frozen tissues and formalin-fixed paraffin-embedded tissues).
