Background
Granulins are a family of secreted, glycosylated peptides that are cleaved from a single precursor protein with 7.5 repeats of a highly conserved 12-cysteine granulin/epithelin motif. The 88 kDa precursor protein, progranulin, is also called proepithelin and PC cell-derived growth factor. Cleavage of the signal peptide produces mature granulin which can be further cleaved into a variety of active, 6 kDa peptides. These smaller cleavage products are named granulin A, granulin B, granulin C, etc. Epithelins 1 and 2 are synonymous with granulins A and B, respectively. Both the peptides and intact granulin protein regulate cell growth. However, different members of the granulin protein family may act as inhibitors, stimulators, or have dual actions on cell growth. Granulin family members are important in normal development, wound healing, and tumorigenesis.

I'd like to get more information about this antibody, which is made by SDIX. We are using it for ICC and get a signal but it looks like it is in the nucleus. GRN hasn't been reported there so we are concerned about the specificity of this antibody. Can you tell me the target epitope region? Do you have sell the target peptide so we can do pre-absorption experiments? Have you tested it in ICC or ICC?
The epitope lies within the Granulin 4 region of the protein P28799 (https://www.uniprot.org/uniprotkb/P28799/entry) (a.a. 281 - 336). Unfortunately we do not sell the target protein or epitope. To my knowledge we do not have any ICC data to aid the client.
Does the antibody bind to a particular part of the granulin protein?
This is made to the middle region of the protein; however, we do not epitope map, so we do not know the specific binding site itself. Since it is polyclonal, it most likely binds to multiple regions.
Does the antibody detect splice variants of granulin?
This antibody targets the region of granulin that is cleaved to produce granulin 4. This region is present in all 3 splice isoforms listed in UNIPROT.
