Western Blot: Glycerol Kinase Antibody [Biotin] [NB120-20600] - Detection of purified Glycerol Kinase under reducing (R) and non-reducing (NR) conditions. Reduced samples of purified protein contained 4% BME and were boiled for 5 minutes. Samples of 1ug of protein per lane were run by SDS-PAGE. Protein was transferred to nitrocellulose and probed with 1:3000 dilution of primary antibody. Detection shown was using Dylight 488 conjugated Donkey anti goat.

Secondary antibody: HRP Streptavidin secondary antibody at 1:40,000 for 30 min at RT. Block: incubated with blocking buffer for 30 min at RT. Predicted/Observed size: 55 kDa, 55 kDa for Glycerol Kinase.

• Application Notes: This product has been tested by ELISA and western blot. This product is assayed against 1.0 ug of Glycerol Kinase in a standard capture ELISA using Peroxidase Conjugated Streptavidin and ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:4,000 to 1:20,000 of the reconstitution concentration is suggested for this product.

Background

Glycerol kinase catalyzes the formation of glycerol 3 phosphate from ATP and glycerol. Dihydroxyacetone and L glyceraldehyde can also act as acceptors; UTP and, in the case of the yeast enzyme, ITP and GTP can act as donors. It provides a way for glycerol derived from fats or glycerides to enter the glycolytic pathway. NB - Glycerol kinase obtained from Cellulomonas spp has an observed molecular weight of 128kDa.
