Western Blot: GAP-43 Antibody [NBP1-41123] - Total protein from Human, Mouse and Rat brain, and SHSY-5Y cells was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-GAP43 in 1% non-fat milk in TBST and detected with an anti-sheep HRP secondary antibody using chemiluminescence.

Background

GAP43 (growth associated protein 43, also called B-50, PP46, calmodulin-binding protein P-57, neuromodulin, neuron growth-associated protein 43, protein F1) is a major component of motile "growth cones" that form the tips of elongating axons as well as the synaptic plasma membranes. GAP43 is associated with nerve growth process and plays a key role in axonal as well as dendritic filopodia induction. It has been detected in a complex containing FGFR4, NCAM1, CDH2, PLCG1, FRS2, SRC, SHC1, GAP43 and CTTN, and binds calmodulin with superior affinity in the absence of Ca2+ than in its presence. Intracellular Ca2+ regulates PKC mediated phosphorylation of GAP43 and affects GAP43's ability to bind calmodulin. GAP43 is integral to growth cone formation, neurite outgrowth, and the development of a functional cerebral cortex. Aberrant expression of GAP43 has been seen in patients diagnosed with schizophrenia and Alzheimer's disease.

   • NBP1-41123 has only been validated in IHC on frozen sections at this time. However, NB300-143 has been validated in IHC on both frozen and paraffin-embedded tissues.
2. Could you tell us the sequence homology between the immunogen of this antibody and the GAP43 protein of canine?
   • The immunogen for NB300-143 shares 82% similarity with canine. NBP1-41123 may be a better choice. It is known to work in dog and in IHC. It has also been published with 10 times in IHC and some of these publications almost certainly used paraffin-embedded tissues.
