Specific for the ~42k - 44k ERK/MAPK phosphorylated at Thr202 and Tyr204. Immunolabeling is blocked by the phosphopeptide used as antigen but not by the corresponding dephosphopeptide. The immunolabeling is completely eliminated by lambda-phosphatase.

The activation of signal transduction pathways by growth factors, hormones and neurotransmitters is mediated through two closely related MAP kinases (p44 and p42), designated ERK 1 (extracellular-signal related kinase 1) and ERK 2, respectively. ERK proteins are regulated by dual phosphorylation at specific tyrosine and threonine sites, which is required for full enzymatic activation. Once activated, MAP kinases phosphorylate downstream components on serine and threonine. Upstream MAP kinase regulators include MEK and Raf-1.
